What is linearity in HPLC and how will you test?
Answer / kishor solanke (alkem)
To check the area(response) of analylite in the sample is
linear with respect to it's concentration OR injection
volume. This parameter done by two method:
1) Detector linearity:
In this linearity test the solution of different
concentration (linear conc.)are injected in triplicate of
each solution & linearity of each concentration calculated
as co-relation coefficient by graphical pattern in excle
against mean srea obtained.
2)Injector Linearity:
In this test the soluiton of same concentration injected in
triplicate with different injection (linear)volumes &
linearity calculated as above.
