5 replicates of standard if %rsd is 2.0% orless
6 replicates of standard if %rsd is more than 2.0%

Yes, it is necessary to inject 6 injection it show your system repeatability (precision), if it is within limit, it means your system is ready to proceed.

As per USP we can inject 3 to 6 , but %RSD vary as per number of injections
