The thing is u should not use mobile phase pH 5.00. any pH
u can use generally +/- 1.00 unit is preferable. in that
also selection of lowe or higher pH is depending on your
compound nature and noise. in whis pH u have less noise
choose that.

u should take pka = +/-2 (ph)
ex; pka 5 ph should be either 3 or 7

u can not say that if Pka is 5 so use buffer near about pka
5 but depends upon the nature of compound , drift and noise.
genrally u can selet pH +/- 1.5 unit of the Pka value of
the compound.
Most important thing is that its not react with buffer,

Compound pka value is 5 u have to choose pka value 5
buffer and adjust ph + or - 1.5 units and buffer uv cut off
should be below 200 nm,good solubulity with organic
solvents.

compound nearly containing buffer will be taken like
ammonium acetate pka 4.8 adjusted ph plus or minus 2
