can we assign roles to user without user master comparison

Without user master comparison, the generated profile is not entered in the user master record. So the user does not access the role or tcodes.
So you must do the user master comparison.

Assignment of roles do not require User Master Comparison. However if the user needs to execute the authorizations he/she have via the above role assignment, User Master Comparison needs to be performed, to have the authorizations of the role reflected in the User Buffer.
