Can you please tell me what and how to test in a web based
application manually? And i manually need to do usability
testing and functionality testing for the same (web based
application. So please tell me how and what to check the
functionality and usability of the web based application?

usability defects are (1)To check Tab Order (2)Alignment
issue (3) Spelling mistake and more.So you can easily check
this defects in the screen.
Functionality defects are validation defects and more.
validation defects's one example: If there is a
textbox.According to business,this textbox should take 10
characters.Then u have to enter more than and equal number
of characters in this textbox.Here u have to check whether
it is taking 10 characters or more than 10 characters.If it
is taking more than 10 characters.Then it is a bug.It is so
easiy to test.
