For Costing Labour Time in Oracle Project Costing you will need to make sure that following setup is done.
1.
You have defined Labour Cost Rate Schedule for the Organization / Operating Unit ( cost rate schedule can be defined at various level of the organization Hierarchy )
2.
You have defined cost rates for each of the Job that is assigned to Employee Assignment. ( I am assuming you are defining cost rate for Job).
It is important that you have got Job associated with employees on the cost rate schedule.
3.
For the Cost rate defined above, you have associated Organization and Cost Rate schedule in Organization Labour Costing Rule.
4.
If you are doing burdening, you need to define burden schedule and all associated setup for Burden schedule ( like, burden cost code, Burden cost base, burden structure ...etc ) and burden schedule should be compiled

Depends on the Costing transaction that you are going to make. Also depends upon the Accounting generation as to where is it going to take place(either in Project Costing or respective subledgers).
Answer 1 is specific and correct to a point when you are decided where the Costing transaction is going to take place.
