Blotting techniques show many limitations in comparison to other molecular techniques. It depends what you're attempting to look for, it's very situational.
- They take up alot of time
- They degrade protein tertiary structure
- They show limited quantitative abilities
- They show lower specificity and sensitivity compared to other techniques.
If you're looking for gene expression I would say reverse-transcriptase PCR is a more appropriate alternative.
