sterilizatio in autoclave is complete only when each and every part of the autoclave reaches 121 degree centigrade.this happens when air in the autoclave is completely replaced by the steam.Air is completely let outside when temperature in autoclave reaches 100 degree centigrade by opening vent for one minute.
