how calculate column dead volume in hplc ?
Answer Posted / subash
we know that K= tr-to/to......1
to=vm/F
where Vm=Volume of mobile phase inside the column
F= Flowrate
Vm= 0.5Xsuare of col diameter(cm)X column lenght(cm)
For a 150mm*4.6mm
Vm=0.5*0.2116*15
Vm=1.587 mL
