If it is possible to test 5000 vuser through single machine? Then how you can test? plz explain

Yes, it is possible, but you need following items,
1) 5000 vusers licensed version
2) The system configuration should be good, like p4 or more
3) Proper virtual user script,
If you have all the above requirement you can do the
test with 5000 usrs
