I have given the protocol for the cyclodextrin glygosyl transferase assay: One ml of appropriately diluted enzyme sample was incubated at 60 °C for 15 min with 5 ml of 1% (w/v) gelatinized soluble starch in 50 mM, 7.0-pH Tris–HCl buffer. Reaction was terminated by boiling the reaction mixture for 3 min and reaction volume was made to 10 ml with distilled water. Two ml of above reaction mixture was withdrawn and mixed with 3 ml of Tris–HCl buffer, 5 ml of 125 mM Na2CO3, and 0.5 ml of phenolphthalein (25 mg phenolphthalein/100 ml absolute alcohol). Absorbance was measured at 550 nm. The percent decrease of sample was calculated with respect to control containing 5 ml of buffer, 5 ml of sodium carbonate and 0.5 ml of phenolphthalein. where Acontrol = absorbance of control and Atest = absorbance of sample. The amount of β-cyclodextrin (β-CD) produced was estimated from the standard graph of 0–500 μg/ml β-CD concentration against % decrease in absorbance. X is strong but has a very low density (1% of traditional earth materials.) and hence light weight. It is a recyclable material. The compression behaviour of X is strain rate dependent. Higher strain rates result in higher initial modulus and higher compression strength. It can also withstand unlimited number of cycling loading provided the repetitive loads are kept below 80% of the compressive strength. The internal structure of the material includes air-traps which make it poor heat conductor. X is non- biodegradable and chemically inert in both soil and water. Most acids and their water solutions do not attack it; however strong oxidizing acids do. Solvents which attack X include esters, ketones, ethers, aromatic and aliphatic hydrocarbons and their emulsions, among others. It does not support bacterial/fungal growth as well .It also has significant acoustic properties and effectively reduces the transmission of airborne sound. X is combustible and should not be exposed to open flame or other ignition sources. Combustion products are carbon monoxide, carbon dioxide, water and soot. Long-term exposure to sunlight causes yellowing and a slight embrittlement of the surface due to ultraviolet light. X is able to withstand the rigours of temperature cycling, assuring long-term performance.

We are using Rosemount 5400 series Radar Level Transmitters in all types of Sump Applications having oily water hdrocarbon petrochemical nature.
