Gas chramatography consist of Gas solid chramatography(GLC)
and Gas liquied chramatography (GLS). In both cases Gas are
used as Mobile phase and either Liquieds are solieds are
act as Statinary phase.
GAS SOLID CHRAMATOGRAPHY :Gas solid chramatography(GLC)not
widely used in pharamacitricul industery because of limited
number of statinary phases are available.The separation
principle is the adsorption in some cases Gas solid
chramatography is used if the solute is not soluble in
stationary phase that situations Gas solid chramatography
used
GAS LIQUIED CHRAMATOGRAPHY:Gas liquied chramatography(GLS)
Gas are used as Mobile phase Liquieds are act as Statinary
phase.Liquieds are coated on the solid support in the form
layer.the layer should be 0.5 to 1.0u
In gas chramatography process the components are convarted
into vapour stage and mixed with gaseous mobile phase.the
compound which is more soluble in stationary phase travels
slower and elluted later if the solute less soluble in
stationary phase travels faster and elluted out first. no
compounds have similar partition coefficient for fixed
combination of stationary phase, Mobile phase and other
conditions. Hence the compounds are separated accourding to
the partition coefficient

GC may b gas solid chromatography or gas liquid
chromatography.In GSC stationary phase used is a solid and
mobile phase is carrier gas .the principle involved is
adsorption
.Whereas in GLC stationary phase used is liquid coated onto
a solid support.and mobile phase is carrier gas .here the
principle involved is partition.based on partition
coefficent,the compound which is more soluble in mobile
phase elutes first and compound which is more soluble in
stationary phase elutes last.carrier gas used is
helium,nitrogen,or hydrogen.GC requires temperature
programming.columns used may be packed or capilllary columns
which may be open tubular columns(SCOT,WCOT).IT also
requires flow regulators,flow
metrs,preheater,detectors(electron capture,thermal
conductivity,flame photometric,flame ionisation,)
