The primary antibody bound to theβ-gal protein on the
membrane. Then a secondary antibody that could recognize
the rabbit antibody was used.
This secondary antibody was attached to theenzyme alkaline
phosphatase (AP). The secondary antibody bound to the
primary antibody andlocalized AP to the band that had β-gal
protein. A substrate for AP was then added.
When this substrate was cleaved by AP, a purple color
appeared.
