AFTER COMPLECTION MEDIA STERILIZATION IN FERMENTER CT WATER

The situation depends upon size of the fermenter and other factors. Holding of media at 121degrees C is a requirement of sterilization to nullify the bioload in the media. Holding of media at high temparature for longer period may degrade nutrients.
1. so we have to bring down the temparature as early possible with out damaging the reactor with considerable positive pressure in the reactor.
2. For bigger fermenters, chilled water usage for bringing down temp from 120degrees is not econamical. so we can use cooling tower water to bring down temp up to 50degrees and then followed by chilled water circulation untill temp reaches to process requirement.
where as for tiny insitu fermenters, it doesnt show any difference.Initially air cool to 100 degrees with slight opening of exhaust valve by maining positive pressure with sterile air passage through sparger. then reduce the temp with chilled water circulation.
