The only difference between manual and automation testing is
- manual testing is performed by physically running the test
cases where in automation test case runes by a automation tool.

There are limitations in both manual an automation say for
instance

one cannot run stress testing or load testing for web
application in manual teasing, practically it is not possible

where in automation one cannot test the font size and
background color test.

financial application involving security concern cannot be
tested automatically as most of the companies prefer manual
testing.

Above are just few examples to prove the limitations
