Plasmid is present in the bacterial cell in the three
distinct forms.
1. Supercoiled
2. Nicked
3. Linear
Through the agarose gel the Supercoiled form move
Quickest.

In order, from slowest running to fastest running, they are relaxed or nicked circular (supercoling not yet accomplished [relaxed] or removed [nicked, a single stranded break]), linear (double stranded break), and supercoiled.

There are 3 conformation observed on 1% agarose gel run of isolated Plasmid DNA, they are -- supercoiled circular DNA, nicked supercoiled circular DNA, and relaxed circular Dna. If the isolated plasmid DNA is not treated with Rnase enzyme then another band is observed in agarose gel known to be DNA masked with RNA. Read lehninger and stryer.
