They are all Human Embryonic Kidney cells (well they are supposed to be, but there is some dispute over their actual cell type with some suggestions they may be neuronal). HEK293 are the original cell line derived and are used for the easy transfectability. HEK293T cells have been transduced with the SV40 (virus) large T antigen, which permits the amplification of plasmids containing the SV40 origin of replication. Thus even if only one plasmid makes it into these cells, many copies can be made permitting both higher levels of expression and longer retention of the plasmid following multiple rounds of mitosis.
